Protein Expression

Recombinant protein production is one of the most powerful techniques used in the Life Sciences. The ability to produce and purify an abundance of the target recombinant protein enables a wide range of possibilities including, its use to diagnose or disease treatment or its use in industrial processes.

At first glance, recombinant protein expression looks straight forward ̶ DNA encoding a desired protein is cloned downstream of a promoter in an expression vector. This clone is introduced into a host cell and the cell’s protein synthesis machinery produces the desired protein. However, in practice protein expression can be very challenging because so many factors may influence the process. For example, some proteins may be subject to protease activity, which may also be addressed by the choice of expression host. Some proteins may have an activity that is detrimental to the host.

What is the Difference Between ScarabXpress-1 & ScarabExpress-2Δ
ScarabXpress1 consists of the ScarabXpress® T7 lac host + a vector containing a T7 promoter such as the pET vectors. ScarabXpress2 consists of Scarab’s pSX2 expression vector + ANY Clean Genome® E. coli host strain.

ScarabXpress2 generally provides higher expression levels of any given target protein. It enables tighter expression control, which is very useful for periplasmic expressed targets and “toxic” targets that are detrimental to the host. In a side by side comparison using the same test protein, ScarabXpress2 produced 29 - 36x more test protein than BL21(DE3) (Figures 1, Figure 2) and a 3000-Fold Induction Range (Figure 3), where as ScarabXpress1 only produced 12x more test protein than BL21(DE3) (Figure 4).
